IMPROVED METHOD FOR DECONTAMINATING BIOLOGICAL MATERIAL BY SEPARATION AND INACTIVATION Russian patent published in 2022 - IPC A61L2/18 C07K14/59 C07K14/765

Abstract RU 2769062 C2

FIELD: separating.

SUBSTANCE: invention relates to a method for decontaminating biological material by applying separation and inactivation techniques, including centrifugation of a three-layer liquid system (a-b-c) wherein the layers are arranged one above the other and differ in the properties of density and/or miscibility; the upper layer (a) comprises an aqueous solution of the biological material in need of disinfection; the lower layer (c) comprises a solution inactivating said viruses, prions and/or bacteria; the intermediate layer (b) located between said upper and lower layers separates said layers and protects said biological material from the impact of the inactivating solution. Said layers therein have different densities, in the order: (a)<(b)<(c), layers (a) and (b) have a density difference in the range from 0.030 to 0.3 g/ml; layer (b) does not mix with layer (a); layer (b) constitutes a mixture of a halogenated solvent and an aromatic organic solvent in a volumetric ratio from 1:0.5 to 1:4, preferably from 1:0.8 to 1:3; layer (c) constitutes a urea solution with a molarity in the range from 7 M to 9 M and a pH from 8.0 to 11.0, or a solution of NaOH with a molarity in the range from 1 M to 2 M.

EFFECT: decontamination of the biological material from viral, prion, and/or bacterial contaminants by separating the contaminants during centrifugation in a three-layer liquid system from the upper layer of a solution of the biological material through an intermediate layer into the lower inactivating layer.
