FIELD: agricultural biotechnology.
SUBSTANCE: method for creating double cabbage haploids (Brassica oleracea L.) in isolated microspore culture refers to the field of agricultural biotechnology and can be used to reduce the labor intensity of obtaining double cabbage haploids in isolated microspore culture. A method characterized in that the isolation and purification of microspores is carried out using a 13% sucrose solution (pH 5.8), initiating stress is exerted by incubation of the isolated and purified microspores in a 13% sucrose solution (pH 5.8) in the dark at a temperature of 32.5±0.1°C for 48 hours, after exposure to initiating stress, the sucrose solution is replaced with a nutrient medium NLN-13 (pH 5.8), then the density of the microspore suspension is determined, adjusted to 4×104 microspores/ml and incubated until the formation of embryoids in a climate room.
EFFECT: invention makes it possible to reduce the laboriousness of obtaining double cabbage haploids while maintaining the frequency of microspore embryogenesis at the level of a standard protocol.
