FIELD: medicine.

SUBSTANCE: invention relates to medical microbiology. Described is a method for producing an antigenic conjugated substance of a haemophilic type b microbe for creating a haemophilic type b conjugate vaccine (Hib), as well as combined vaccines containing ADS + Hib, DTaP + Hib. Haemophilus influenzae type b (Hib) No. B-7519 strain is cultured on chocolate agar with addition of bacitracin and subsequent transfer to a liquid nutrient medium of the following composition: hydrochloric acid hydrolyzate of casein, glucose, yeast extract, nicotinamide dinucleotide (NAD), hemin, group B vitamins, NaHPO4, KH2PO4. Culture is inactivated by treating it with 0.1 % formaldehyde. Further, Hib cells are separated from the culture fluid by filtration on plate membrane filters in a tangential flow. When isolating capsular polysaccharide (CP) Hib, cetyltrimethylammonium bromide (cetavlone) is used, wherein the amount of cetavlone is calculated taking into account the content of CP in the concentrate by formula: mC = Px48, where mC is the amount of cetavlone, Ρ is the amount of phosphorus in the volume of the ultrafiltrate concentrate, 48 is the conversion factor. CP is cleaned by hot phenol treatment with subsequent alcohol deposition. For the conjugation step, tetanus anatoxin is used, activating it by a carbodiimide method, attaching a linker molecule ADH (adipic acid dihydrazide) to it. Conjugation is carried out by adding to activated tetanus toxoid purified capsular polysaccharide Hib treated with a cyanylating agent.

EFFECT: invention enables to create an antigenic conjugated substance of a haemophilic type b microbe for producing a haemophilic type b conjugate vaccine (Hib), as well as combined vaccines containing ADS + Hib, DTaP + Hib.
