SUBSTANCE: invention relates to medicine, namely ophthalmology. The separation of the pre-Descemet layer, Descemet membrane and endothelium from the residual stroma of the donor tissue is carried out using the large bubble technique. The corneoscleral disc is placed with the endothelium upwards, a 30 G needle connected to a 5 ml syringe filled with air from the side of the scleral ring is inserted into the middle layers of the stroma until a large type 1 bubble is formed. Paracentesis of the bubble is performed, trypan blue dye is injected and a detached graft is carved around the bubble circle. The corneoscleral disc is placed and fixed in the bed of the plastic base of the vacuum chisel with the endothelium up, and the type 1 bubble is formed using a syringe having a piston with a spring and connected to a needle.

EFFECT: method provides stable fixation of the donor tissue when the needle is inserted, reduces intraoperative risks of bubble rupture, and preserves the donor material.
