FIELD: biotechnology.

SUBSTANCE: method for the purification of full capsids of an adeno-associated virus (hereinafter – AAV) from a concentrated AAV fraction containing empty AAV capsids and full AAV capsids provides for: (i) loading to a zonal rotor of a) the concentrated AAV fraction and b) at least two sugar solutions, each of which is characterized by different sugar concentration, and each of which contains sugar at a concentration equivalent to sucrose concentration in the range from about 45% (in mass terms) to about 65% (in mass terms) of sucrose, (ii) operation of an ultracentrifuge containing a zonal rotor in a periodic mode, after which sugar gradient is formed, (iii) production of a sugar gradient fraction for the production of AAV fraction, wherein at least or about 60% of AAV particles in AAV fraction are full AAV capsids, wherein the total volume of sugar solutions and the concentrated AAV fraction is less than or equal to the volume of the zonal rotor, and a ratio of the volume of sugar solutions to the volume of the concentrated AAV fraction is less than or equal to one.

EFFECT: method allows for obtaining a high-purity effective product suitable for clinical use.
