SUBSTANCE: invention relates to the field of biotechnology, in particular to a method for the modification of a target locus of interest, including the delivery to the specified locus of a composition not found in nature or constructed, including monomolecular effector protein CRISPR-Cas containing three catalytic domains of RuvC nuclease, but not containing HNH domain, and one or more components, which are nucleic acids including a heterologous directing sequence and tracr-RNA sequence. A composition not found in nature or constructed, and a system for the modification of a target sequence of interest, as well as a delivery system containing it, and a vector system are also disclosed. The invention also relates to a particle, a cell, a cell line containing it, as well as to a method for the production of a plant, and a method for the inhibition of cell growth.
