FIELD: biotechnology.

SUBSTANCE: invention relates to biotechnology, in particular, the method involves three consecutive rounds of PCR with a combinatorial library and a modified dNTP candidate for SELEX. The conclusion about the applicability of a specific dNTP derivative is based on the nature of the change in the signal accumulation curve during three rounds of real-time PCR and does not require rounds of aptamer selection. If the library degenerates (becomes less representative) during amplification, then the particular dNTP modification cannot be used with the selected polymerase and other selected library amplification conditions, as it results in competitive amplification. In the case when the nature of the signal accumulation curve does not change, it is concluded that the modified deoxynucleoside triphosphate used does not affect the distribution of oligonucleotides with different sequences in the library, i.e. does not lead to a change in its composition from the point of view of the detection method used.

EFFECT: method is applicable for rapid assessment of the substrate suitability of modified dNTPs for mod-SELEX and will be useful in the selection of aptamers for clinical diagnostics, medicine and research.
