FIELD: biotechnology.
SUBSTANCE: invention relates to the field of biotechnology, in particular to the cultivation of plant tissues and organs, and can be used in agriculture and forestry to obtain high-quality planting material. The method for sterilization of birch explants in vitro using nanoparticles of copper oxide is carried out as follows. Young birch shoots are pre-washed in water with the addition of surfactants (surfactants) - potassium stearate, cut into segments 3-5 cm long, and then sterilized for 35 minutes in a solution consisting of 200 ml of distilled water and 200 mcl of 5% solution of sodium hypochlorite, followed by washing in distilled water. The main sterilization of birch shoots is carried out in a laminar box in a solution containing 15 ml of a 5% sodium hypochlorite solution and 85 ml of sterile distilled water for 15 minutes. Subsequent rinsing is also carried out with sterile water. Sterile shoots of birch are cut under aseptic conditions into segments 1.5-2 cm long with one axillary bud - explants, which are subsequently planted on a WPM nutrient medium optimized for introducing birch explants into in vitro culture and containing spherical copper oxide nanoparticles with a diameter of 30-80 nm at a concentration of 0.1 mg/l.
EFFECT: method for in vitro sterilization of birch explants using copper oxide nanoparticles is characterized by increased safety and efficiency compared to known methods and makes it possible to prolong the sanitizing effect of copper oxide nanoparticles in the WPM modified nutrient medium when introduced into an in vitro culture, as well as to reduce the production time and increase the amount sterile explants.
