FIELD: biotechnology.
SUBSTANCE: invention relates to the field of biotechnology and concerns methods for measuring the local temperature of the medium, in particular intracellular temperature. A new approach to measuring intracellular temperature with high accuracy (from 0.1 to 0.3°C) and the possibility of measuring with high spatial resolution (up to 300 nm) in biological media (cells and tissues), as well as the temperature of cells and tissues using optical methods is proposed. To measure temperature, it is proposed to use the phenomenon of heterogeneity of spectral-temporal characteristics of anti-Stokes fluorescence of phycobiliproteins, namely, registration of the kinetics of attenuation of anti-Stokes fluorescence of phycobiliproteins in cells and tissues.
EFFECT: the invention provides the possibility of functional visualization of the local temperature of cells and tissues by using the phenomenon of conformational mobility of chromophores of phycobiliproteins, whose fluorescence lifetime when excited by infrared light from 750 to 790 nm depends on the local temperature with high accuracy.
