SUBSTANCE: invention relates to the field of biotechnology. The invention is a test system for detecting DNA of the bovine leukosis provirus (Bovine leukosis virus, BLV) in food by real-time polymerase chain reaction, including plastic vials and test tubes, a thermostable Tag-polymerase enzyme, a buffer for setting up the reaction, a mixture of four deoxynucleotide triphosphates, an internal exogenous control sample in the form of a suspension of T4 bacteriophage with a concentration of 5×103 copies of nucleotide sequences per 1 μl, a positive control sample, which is a mixture of recombinant plasmid DNA containing a fragment of the Bovine leukosis virus (BLV) genome) and a fragment of the T4 bacteriophage genome specific for the Bovine leukosis virus (BLV) provirus genome and the T4 bacteriophage genome, as well as oligonucleotide primers, probes with the following nucleotide sequences:
at the same time, for a positive control sample, a fragment of the bovine beta-actin gene is additionally used as an endogenous internal control with primers having the following nucleotide sequences:
in this case, all fragments of the genomes and the beta-actin gene of cattle included in the mixture of the positive control sample were taken in equal parts.
EFFECT: invention makes it possible to reliably diagnose the detection of the bovine leukosis virus (BLV) provirus genome in food products and in raw materials for their preparation of animal origin.
