FIELD: biotechnology.
SUBSTANCE: invention relates to biotechnology and constitutes a recombinant plasmid DNA pSp-raPLY containing a gene for the expression of recombinant atoxic pneumolysin S. pneumoniae, the expression whereof is controlled by the promoter of phage T5, two lactose operons, lambda t0 of the transcription stop region, transcription terminator rrnBT1. The pSp-raPLY plasmid contains the ampicillin resistance gene, a ColE1 plasmid replication initiation site. Said plasmid is used to transform cells of the recipient strain of Escherichia coli M15. The recipient strain of E. coli M15 contains the pREP4 plasmid including the gene of a lac protein binding to lac operons and the kanamycin resistance gene. The genome of the strain of E. coli M15 contains a highly expressed gene of the RNA polymerase of bacteriophage T5.
EFFECT: strain of E. coli M15/pSp-raPLY with a productivity of at least 5 mg of the recombinant atoxic form of pneumolysin S. pneumoniae per 1 litre of culture medium or 5 to 10 g wet biomass of strain cells.
