METHOD FOR DETECTING THE CAUSATIVE AGENT OF PSEUDOTUBERCULOSIS Russian patent published in 2022 - IPC G01N33/52 A61B5/00

Abstract RU 2783710 C1

FIELD: laboratory diagnostics and immunology.

SUBSTANCE: invention relates to laboratory diagnostics and immunology, and can be used to detect the causative agent of pseudotuberculosis. The method includes: applying the test material to a substrate with a pore size of 0.17-0.45 μm, on which the test material is applied pointwise in a volume of 1-2 μl, after complete absorption of the test material, the substrate is washed twice with saline, then placed in a 2% solution bovine serum albumin in physiological saline for 15 minutes, then the substrate is washed twice with saline, once with distilled water and placed in a solution of immunoglobulin G isolated from agglutinating pseudotuberculous serum, labeled with particles of colloidal silver 5-9 nm in size, with exposure for 40 minutes. After that, the substrate is washed twice with saline and once with distilled water, then it is immersed for 3 minutes in the developer, while the developer is an aqueous solution consisting of 0.5% citric acid, 0.1% metol and 0.2% nitric acid and then washed with running water. The presence of the causative agent of pseudotuberculosis in the test material is visually determined. If gray stains from dark gray to light gray are formed on the substrate at the places where the material is applied, then the test material contains the causative agent of pseudotuberculosis, if there is no staining on the substrate, the material does not contain the causative agent of pseudotuberculosis.

EFFECT: method makes it possible to detect minimal amounts of the pseudotuberculosis pathogen, Expressen (total analysis time is 1 h 10 min), economical, easy to set up and take into account the results of the reaction, available for wide use, including in the field, does not require expensive reagents, equipment and specially trained personnel.
