FIELD: biotechnology.
SUBSTANCE: invention relates to the field of biotechnology, specifically to the production of proteins from blood serum, and can be used to isolate and purify alpha-2-macroglobulin (alpha-2-MG). A method for isolating and purifying alpha-2-MG is proposed, including protein fractionation with ammonium sulfate 40% saturation, gel filtration, and ion-exchange chromatography. The blood serum of healthy men is used as the starting material, gel filtration is carried out on Sephadex G-200 in 0.1 M sodium phosphate buffer, pH 7.3 at a ratio of the height of the hydraulic column to the height of the working part of the column - 1/5 and speed elution - 15-25 ml / cm2⋅h, the final stage of purification is carried out by ion-exchange chromatography on QAE-sephadex A-50 in 0.05 M Tris-HCl buffer with pH-6.2, and the elution of alpha-2-MG is carried out in a single-stage ionic strength gradient up to 0.45 mol/l.
EFFECT: increased efficiency of isolation and purification of alpha-2-MG.
