SUBSTANCE: invention relates to the field of analytical chemistry. A method for the quantitative determination of malondialdehyde in a cell culture medium, which is a Hanks solution after incubation with Caco-2 cells for 3 hours, is disclosed, including the use of the HPLC-MS/MS method, characterized by sample preparation by mixing 10 μl of a cell culture medium sample with 90 µl of acetonitrile, followed by shaking for 10 min and further centrifugation of the sample at 10000g for 10 min at a temperature of +4°C, followed by injection of 5 µl of the sample using an autosampler and chromatography on a column UCT Selectra C18 4.6 mm×100 mm, 3um, 100 A with Selectra C18 Guard Cartridges SLC-18GDC46-3UM at column temperature 35°C, isocratic elution at a flow rate of 300 µl/min, using a mobile phase consisting of 20% acetonitrile and 80% aqueous ammonium formate with a concentration of 10 mmol / l, while ionization is carried out by forming an electrospray in a negative mode at atmospheric pressure, using 2700 V electrospray voltage, sheath gas 50 arb, auxiliary gas 10 arb, purge gas 1 arb, evaporator temperature 350°C, ion transport tube 300°C and use MRM mode m/z 71.1 Da → 41 Da at argon pressure 1 mTorr.
EFFECT: invention provides for the development of a sensitive, selective, accurate, precise method for the quantitative determination of MDA in a cell culture medium.
