FIELD: biomedical sciences.

SUBSTANCE: invention relates to cell biology, cytology, histology, and is intended for simultaneous visualization of a mast cell population with signs of metachromasia and tryptase-positive granules in histological sections of tissue micropreparations. The method includes the following: fixation with 10% neutral formalin, immunohistochemical detection of tryptase using monoclonal antibodies, with completion of preparation by placing the micropreparation in the mounting medium. After immunohistochemical staining, immediately, without allowing the micropreparation to dry out, 1.5–2 ml of toluidine blue solution is applied and afterwards incubation for 3 to 10 minutes is required. Rinse with isopropyl alcohol in shifts in two portions of isopropyl alcohol for at least 30 seconds in each portion; clarify at least three times in O-Xylene, keeping each time for three minutes.

EFFECT: method makes it possible to increase the accuracy of assessing the protease profile of a mast cell population, taking into account the cytotopography of tryptase-containing granules and their directed migration from the nucleus to the plasma membrane using bright-field microscopy, to objectively interpret and characterize the involvement of MC during adaptive or pathological processes, and also to visualize the process of biogenesis and secretory pathways of TK tryptase, cytotopography of its granules and their directed migration from the nucleus to the plasma membrane.
