FIELD: medical diagnostics of diseases of the cardiovascular system.

SUBSTANCE: method of detecting mast cells on histological preparations of the human heart includes fixing the preparation in 10% formalin, dehydration, paraffin embedding, preparation of histological sections with a thickness of 5 to 10 μm, applying them to glass slides, drying in a thermostat, deparaffinization in xylene, rehydration, staining in a freshly prepared working dye solution, washing of sections and tinting of preparations with nuclear strong red, dehydration in alcohols of ascending strength, clarification in xylene and inclusion in a fixing medium. The preparations are subjected to progressive staining with alcian blue 8 GX (CI74240) at a temperature of 40°C for 18–20 hours, and the working solution of alcian blue dye is obtained by mixing 1% alcian blue and 5% acetic acid in a ratio of 1:40, after which the color is fixed for 30 s in 0.01% ammonia solution, then the cell nuclei are stained for 1 min with a 0.2% acidic solution of nuclear strong red (CI60760), which is prepared by dissolving 10 g of ammonium alum in 100 ml of distilled water, followed by the addition of 0.2 g of nuclear strong dye red (CI60760), heating to boiling, cooling and adding 5 ml of glacial acetic acid.

EFFECT: method is easy to reproduce, can be used to study archival material, fixed in 10% formalin, stored in paraffin blocks.
