METHOD FOR STAINING TRYPTASE-POSITIVE MAST CELLS IN TISSUE SLIDES WITH COUNTERSTAINING USING MAY-GRÜNWALD SOLUTION Russian patent published in 2022 - IPC G01N1/28 G01N1/30 G01N33/53 G01N33/577

Abstract RU 2781558 C1

FIELD: medicine.

SUBSTANCE: invention relates to medicine, namely, to cellular biology, cytology, and histology, and can be used for staining tryptase-positive mast cells in tissue slides with counterstaining using May-Grünwald solution. The sections undergo deparaffinisation and staining by the immunohistochemical method using monoclonal antibodies, applying secondary antibodies detected by the reagent, followed by histochemical staining using May-Grünwald solution. After immunohistochemical staining, without letting the slide dry, 1.5–2 ml May-Grünwald solution is applied to the slide and left for 5 minutes, followed by submerging the slide at least 5 times in a 0.4–0.8% solution of glacial acetic acid.

EFFECT: method allows identification of the general population of mast cells in bright-field microscopy with simultaneous immunohistochemical detection of tryptase-positive mast cells and objective characteristic of the functional activity of the mast cells, due to the objective evaluation of the level of expression of tryptase in the population of mast cell by means of the claimed method for staining tissues.
