FIELD: cell biology; medicine.
SUBSTANCE: invention relates to a method for obtaining cells derived from the dental pulp, enriched with pluripotent stem cells. To implement this method, first, the stage of hydrolysis of the dental pulp with collagenase and neutral metalloprotease is carried out to prepare a suspension of the dental pulp. This suspension is then cultured to proliferate the pluripotent stem cells contained in the suspension at 37°C in the presence of 5% CO2 in DMEM supplemented with fetal calf serum at a concentration of 20% (v/v) and antibiotics, changing the medium every 2–4 days. Thereafter, the proliferated pluripotent stem cells are frozen in a state in which the pluripotent stem cells are suspended in the first cryopreservation liquid. The frozen pluripotent stem cells are then thawed. Thereafter, they are cultured in a state in which the pluripotent stem cells are attached to the surfaces of the carrier particles for the proliferation of the pluripotent stem cells on the surface of the carrier particles at 37°C in DMEM supplemented with 10% (v/v) fetal calf serum supplemented with glucose. Then, the step of bringing the carrier particles into contact with trypsin is carried out to separate the pluripotent stem cells attached to the surfaces of the carrier particles from the carrier particles, and the process is completed by the step of preparing a suspension of the separated pluripotent stem cells.
EFFECT: invention improves the efficiency of obtaining pluripotent stem cells from dental pulp and provides a novel preparation of pluripotent stem cells in a form that can maintain stability suitable for distribution and storage.
