FIELD: allergology; immunology.
SUBSTANCE: invention relates to a method of differentiating M. avium supsp. paratuberculosis from other types of mycobacteria. Mycobacteria are injected subcutaneously into guinea pigs, the sensitizing properties of mycobacteria are evaluated after 30 days by the method of a simultaneous test with PPD for mammals, by intradermal injection into the lateral surface of the abdomen — on the right, and an allergen designed to differentiate nonspecific reactions to tuberculin, into the lateral surface of the abdomen — on the left, the intensity of allergic reactions is determined after 24 hours and the intensity of these reactions to both allergens are compared. Subcutaneous injection of cultures of mycobacteria is carried out at four points in the withers and sacrum at a dose of 5 mg as part of an adjuvant including 15 parts of lanolin and 85 parts of vaseline oil in a volume of 0.5 ml per animal. As an allergen intended for differentiation of non-specific reactions to tuberculin, the Paraavium complex allergen is used consisting of tuberculoproteins of the avian mycobacterium strain M. avium 22-82 and tuberculoproteins of the strain M. avium supspecies paratuberculosis 19698 ATCC in equal volumes 50:50 in equivalent activity with total protein content — 1 mg of protein in 2 ml of solvent. Differentiation is carried out according to the diameter of the swelling — a greater increase in the diameter of the swelling on "Paraavium" than on the PPD for mammals indicates sensitization of guinea pigs by paratuberculosis mycobacteria.
EFFECT: invention provides differentiation of M. avium supsp. paratuberculosis from other types of mycobacteria.
