FIELD: genetic engineering; biotechnology.
SUBSTANCE: guide RNA molecule of the CRISPR-Cas system, a ribonucleoprotein complex of the CRISPR-Cas system for detecting hepatitis C virus RNA, and kits for detecting hepatitis C virus RNA are proposed. A guide RNA molecule of the CRISPR-Cas system can bind to targeted highly conserved genome sites of hepatitis C virus and has a consequence selected from SEQ ID NO: a sequence selected from SEQ ID NO: 1-5. The ribonucleoprotein complex of the CRISPR-Cas system is formed from the RNA-guided DNA endonuclease LbCpf1 from Lachnospiraceae and at least one guide RNA. The kits contain a CRISPR-Cas guide RNA molecule, guided DNA endonuclease LbCpf1 from Lachnospiraceae, instructions for use, and additionally one or more specific oligonucleotides selected from SEQ ID NO: 6-11.
EFFECT: inventions provide ultrasensitive detection of hepatitis C virus RNA of genotypes 1b and 3a up to single (2,3) copies of RNA in one reaction.
