FIELD: veterinary medicine.
SUBSTANCE: invention relates to the pharmaceutical industry and fish industry, and in particular to a method for production of a substance of fat-soluble vitamin D. The claimed method consists in the hydrolysis of raw materials in the presence of potassium hydroxide and ethyl alcohol when heated, where the raw materials are cultivated fish species: chum salmon, pink salmon, coho salmon, trout, flounder, mackerel, as well as non-recyclable fish production waste, which is crushed with a dispersing element with a rotation frequency in the range of 8,000–8,200 rpm for 10 minutes at a temperature of 20–24°C with the supply of inert gas nitrogen or carbon dioxide, the crushed raw materials are homogenized for 30 minutes carried out in sealed bags with purging them for 20–30 seconds with nitrogen or carbon dioxide in a blade-type homogenizer, as well as in carrying out the destruction of the protein part of the raw material in an inert gas environment and continuous stepwise heating in an installation with microwave radiation, which starts from 400 W, gradually increasing to 1,000 W; by adding 12.5% potassium hydroxide solution in an amount of 3.4 ml per 10 g of raw materials, 3.4 g of ascorbic acid and 134 ml of ethyl alcohol 96%, which is carried out for 30 minutes with a constant pressure in the system of at least 3 atm, and the temperature is increased every 10 minutes in increments of 30°C at a temperature range of 20 to 90°C; after 30 minutes, the pressure is released by cooling in the closed state to room temperature, after which the reaction vessels are placed on a shaker for 15 minutes and then the hydrolyzed mixture is diluted with deionized water in an amount of 300 ml and ethyl alcohol in an amount of 67 ml, and fat-soluble vitamins are extracted with petroleum ether or n-hexane for 2 min, after separation of the mixture, the organic phase contains the final product, the extraction is repeated three times with 100 ml portions of the extractant, the extracts are combined, washed from alkali, and to remove water, the resulting extract is filtered through anhydrous sodium sulfate; the solvent is removed in a rotary evaporator at a temperature not exceeding 50°C.
EFFECT: above method provides for the production of a substance using fish industry objects containing fat-soluble vitamin D using low-waste technologies.
