FIELD: food industry; chemical industry.

SUBSTANCE: method of sample preparation for quantitative determination of vitamin A in the form of retinol acetate in range of 500 thousand–1.5 million IU/g or vitamin D3 in the form of cholecalciferol in range of 300–700 thousand IU/g in the analyzed sample by HPLC method contains addition of antioxidant and water-alcohol solution of potassium hydroxide in ethanol with ratio of 2 g KOH: 2 cm3 of water for analysis: 18 cm3 of rectified ethyl alcohol, saponification of the obtained mixture for 30 minutes, followed by cooling to room temperature, adding to the obtained hydrolyzate water, successive triple extraction with hexane with combining the extracts, washing combined hexane extract with distilled water, adding anhydrous sodium sulphate, followed by evaporation, dissolving the obtained dry residue in a liquid, followed by holding in a dark place and filtering before conducting high-performance liquid chromatography, differs by fact that 10% aqueous solution of ascorbic acid is added to a weighed portion of the test sample as an antioxidant, saponification is carried out at temperature of 70°C, after evaporation, the obtained dry residue is dissolved in isopropyl alcohol and subjected to reversed-phase high-performance liquid chromatography.

EFFECT: invention makes it possible to reduce indices of error of procedure to 15%.
