SUBSTANCE: invention relates to a method of dynamic filtration of a cross-linked biopolymer hydrogel to remove unwanted molecules from the gel. A method of dynamic filtration of a cross-linked biopolymer-based hydrogel involves transferring the cross-linked biopolymer-based hydrogel to a dynamic filtration device equipped with one or more semi-permeable filter disks, and diafiltration of the gel. Gel diafiltration involves concentration of the gel by applying rotation speed in the range of 20 to 500 1/min and excess pressure in the range of 0.5 to 6 bar to a given concentration or pumping the gel directly into the working chamber of a dynamic filtration device, performing diafiltration to reduce the amount of unwanted molecules including non-reacting or non-linked polymer cross-linking molecules and their degradation products, by applying a rotation speed in the range of 20 to 500 1/min and an excess pressure in the range of 0.5 to 6 bar. Next, optionally, a mixture containing a non-crosslinked polymer, in particular non-crosslinked hyaluronic acid, and water are added to the gel. The biopolymer-based hydrogel is made from a biodegradable polymer selected from hyaluronic acid, heparosan or their salts or derivatives or combinations thereof. The biopolymer hydrogel is cross-linked by a chemical cross-linker containing at least two groups capable of reacting with one or more functional groups of the biopolymer hydrogel.
EFFECT: invention is aimed at removing unwanted molecules from a gel while maintaining the structural stability of the gel.
