FIELD: medical immunology.
SUBSTANCE: invention relates to a method of predicting the severity of a systemic inflammatory response. A method of predicting the severity of a systemic inflammatory response (SIR) by determining the hyperactivity of the complement system, performing a lysis reaction of group AB erythrocytes [E(AB)] with human serum, characterized by the following: 50% serum, 0.5% E(AB) suspension are used, under VBS2+ buffer conditions at (37.0±0.5)°C, incubated for 10 min, the reaction of complement-dependent lysis of E(AB) is assessed turbidimetrically by the decrease in the optical density of the E(AB) suspension at a wavelength of 620 nm, a calibration graph of the dependence of the optical density A620 on the lysis of human erythrocytes is constructed, which is used to determine the degree of erythrocyte lysis in the test samples, determine the titer of antibodies to antigens of human erythrocytes of the AB group in the hemagglutination test, and calculate the coefficient of complement-activating function of anti-AB antibodies (Kcafa) using the following formula: Kcafa=(U/T)×20, where Y is the degree of lysis, %; T is reverse titer of anti-AB antibodies to human erythrocytes of group AB; 20 is conversion factor in units/ml; with elevated Kcafa (more than 100 units/ml) and hyperactivity of the complement system (more than 60%), a severe course of SVR is predicted.
EFFECT: above method allows to individually predict the severity of Covid-19 with the choice of treatment tactics using specific inhibitors of the complement system.
