FIELD: biotechnology.

SUBSTANCE: method is proposed for determining the activity of the classical and alternative complement pathways in the blood serum of mice, including the lysis reaction of 0.25% suspension of rabbit erythrocytes sensitized with human antibodies (EcrAT), 12.5% mouse serum at 37.0±0.5°C, construction of a calibration graph of the dependence of the optical density of A620 suspension on the lysis of rabbit erythrocytes, which determines the degree of EcrAT lysis. The total complement activity is determined under VBS2+ conditions and incubated for 10 min, the activity of the alternative complement pathway is determined at the same ratios in the hemolytic system only under VBS-Mg2+ -EGT conditions and 30 min incubation; mouse classic complement pathway activity is defined as the difference between total complement activity and that of the alternative complement pathway.

EFFECT: invention provides an extension of the arsenal of methods for studying complement-mediated erythrocyte lysis over time.
