FIELD: biotechnology.
SUBSTANCE: invention relates to a method of obtaining a culture of lymphocytes enriched with tumor-specific clones of T-lymphocytes. The method involves the steps of isolating autologous tumor-infiltrating lymphocytes (TILs) from a patient suffering from solid cancer, autologous TILs are cultured in a medium containing IL-2 and at least one of agents selected from IL-21 and a signal transduction blocking antibody PD-1:PD-L1, determine the repertoire of nucleotide sequences of T-cell receptors using high-throughput sequencing, assess the degree of enrichment of cultured TILs with tumor-specific clones using bioinformatics analysis, which determines the presence of clusters of convergent variants of T-cell receptors in the repertoire.
EFFECT: invention is effective for large-scale expansion using known ex vivo expansion protocols and administration to a cancer patient during adoptive cell therapy.
