FIELD: biotechnology.

SUBSTANCE: method involves antigenic activation of obtained mature DC using a DNA construct encoding epitopes KIFGSLAFL and RLLQETELV of the tumor-associated HER2 protein, co-culturing with autologous mature antigen-activated DC from the adhering fraction of all non-stick MNC fraction cells, followed by isolation of DC and MNC specific to these CTL epitopes from the co-culture by magnetic separation using antigen-specific reagents providing reversible staining of the isolated cells. After isolationn of HER2-specific CTL, their proliferation is stimulated by culturing in the presence of simultaneously introduced cytokines rchIL-2, rchIL-7 and rchIL-15 with the final concentration of 50 ng/ml for each for 14-20 days.

EFFECT: invention provides induction of an effective antigen-specific antitumor cytotoxic immune response against tumor cells with overexpression of the HER2 antigen, characterised by an increase in the percentage of dead HER2-expressing tumor cells.
