FIELD: organic chemistry.
SUBSTANCE: invention relates to the production of oligosaccharides. The proposed method of producing 2'-fucosyllactose using a genetically engineered Escherichia coli cell involves providing a genetically engineered Escherichia coli cell that produces 2'-fucosyllactose. Moreover, the Escherichia coli cell is genetically engineered to express at least one heterologous α-1,3-fucosidase which carries out the intracellular degradation of 3-fucosyllactose and 2'3-difucosyllactose formed during the intracellular biosynthesis of 2'-fucosyllactose. Escherichia coli cell reuses such degradation products as lactose and fucose obtained as a result of the enzymatic activity of the specified α-1,3-fucosidase to produce 2'-fucosyllactose, wherein the genetically engineered Escherichia coli cell contains a bifunctional L-fucokinase/L-fucose-1-phosphate guanylyl transferase to conserve fucose to produce 2'-fucosyllactose. Genetically engineered Escherichia coli cell is cultivated under conditions and in a medium that are permissive with respect to the production of 2'-fucosyllactose, thereby obtaining 2'-fucosyllactose and possibly isolating 2'-fucosyllactose. Also the following is proposed: a genetically engineered Escherichia coli cell for the production of 2'-fucosyllactose and its use for the production of 2'-fucosyllactose.
EFFECT: invention is aimed at producing 2'-fucosyllactose with fewer non-target by-products.
