FIELD: biotechnology.
SUBSTANCE: following is presented: a method of transforming competent cells of bacteria of the genus Lactobacillus with the plasmid pLEM415-ldhL-mRFP1, having the nucleotide sequence SEQ ID NO: 1, containing the erythromycin resistance gene erm, the constitutive promoter of the idhL gene and the coding sequence of the monomeric red fluorescent protein mRFP. After adding the MRS nutrient medium and erythromycin, the bacterial cultures are incubated for 16–18 hours, and the transformation results are detected by analyzing the growth of fluorescence using the FAM channel.
EFFECT: method makes it possible to accelerate the assessment of the effectiveness of different protocols for the preparation of competent cells of bacteria of the genus Lactobacillus, and also makes it possible to simultaneously screen a large number of protocols for the preparation of competent cells, chemicals that increase the efficiency of transformation, and different strains of bacteria of the genus Lactobacillus.
