FIELD: biotechnology.
SUBSTANCE: immunogenic modified F protein of human metapneumovirus (hMPV) or a fragment thereof is described, stabilized in a pre-fusion conformation, containing a recombinant single-chain polypeptide that does not contain a fusion peptide (FP) and contains an F2 domain, a heterologous peptide linker and an F1 domain, with the said linker located between specified F2 and F1 domains and contains a cysteine residue that forms a disulfide bond with a non-natural cysteine residue present in the specified F1 domain. Also the following is presented: an immunogenic composition containing an immunogenic protein or a fragment thereof, additionally containing a pharmaceutically acceptable carrier and/or excipient, wherein the said composition can induce neutralizing antibodies that recognize the native hMPV F protein. Also the following is provided: a method of treating or preventing hMPV infection in a human subject, comprising administering to the human subject an immunogenic composition or vaccine containing an immunogenic protein or fragment thereof.
EFFECT: invention expands the arsenal of means for treating or preventing hMPV infection.
