FIELD: biotechnology.
SUBSTANCE: invention relates to biotechnology, namely to adaptation of triploid aspen (Populus tremula L.) micro-clones to ex vitro conditions for reproduction of wood crops in forestry. Method comprises selecting micro-clones with size of 30 mm, micro-clones are sprayed with Phytolavin VRK — 1 ml/l, cultivated in a climatic chamber with addition of nutrient solution MS with succinic acid — 1 g/l and gibberellic acid — 1 mg/l, Phytolavin VRK — 1 ml/l. At the stage of adaptation and rooting, illumination of 3000 lx, humidity of 90–99% and temperature of 21–23 °C. When additional lateral roots appear and their length increases, moisture content is gradually reduced to 50% by opening ventilation holes of the climatic chamber. After four weeks from the moment of planting, micro-plants are transferred into a non-sterile substrate consisting of peat and agroperlite at ratio of 1:0.5.
EFFECT: invention makes it possible to simplify process of micro-clones adaptation and to increase triploid aspen survival rate to ex vitro conditions.
