FIELD: medicine.

SUBSTANCE: invention refers to diagnostics of spermatogenesis and infertility disorders in urology, andrology, gynecology, reproduction, dermatovenerology, oncology and concerns a method of male reproductive function examination. Method includes: thinning of viscous ejaculate, if viscosity exceeds threshold value of 20 mm; measurement of concentration and total number of sperm cells in ejaculate; staining and determining the concentration of peroxidase-positive cells; staining and determining the proportion of live and/or dead sperm cells; determination of sperm cell motility; determination of aggregation, agglutination; determination of the presence of ejaculate cell elements, such as erythrocytes, lipoid, amyloid bodies, macrophages, spermiophages, spermatogenesis cells. Morphological examination is carried out, during which staining is performed to differentiate the morphology of sperm cells and the proportion of normal and pathological forms is determined. Based on the results of the macroscopic and morphological examination, a spermogram is obtained and semiological diagnoses are made: normozoospermia, oligoasthenoteratozoospermia, asthenozoospermia, oligoasthenozoospermia, dyskinesia, viscosipathy, pyospermia, cryptozoospermia, azoospermia, haemospermia, oligospermia, agglutination, necrozoospermia. During microscopic and morphological examination the following actions are additionally performed: whole portion of the treated ejaculate is transferred into a cone-shaped sterile disposable polypropylene test tube with volume of 10–12 ml; a portion of ejaculate is centrifuged in a medical centrifuge at 1,000 rpm for 15–20 minutes; separating the supernatant without capturing the sediment with a sterile tip using a single-channel variable volume pipette dispenser; 5–10 mcl of the supernatant-free ejaculate sediment is placed on the side of a pre-prepared and marked or barcoded slide with high adhesion to a positively charged surface; slide with deposited ejaculate sediment is placed in device for preparation of standardized smears; ejaculate sediment on a slide is dried in air and stained with dyes according to the Romanovsky hardware method using an automatic device for staining glass preparations; in addition, the following ejaculate parameters are recorded: leukocytes 3 or more in the field of view or COE-pyospermia at a total magnification of x100, macrophages-monocytes 2 or more in the field of view at a total magnification of x100,—multinucleated giant cells, degenerative changes of urothelium, glandular epithelium, flat epithelium with signs of papillomavirus infection of high carcinogenic risk, inclusion in glandular epithelium and/or urothelium, morphologically similar to Cl reticular bodies. trachomatis, glandular epithelial urothelium cells 5 or more in the field of view at a total magnification of x100, polymorphic microbiota, Candida fungal elements, such as spores, pseudomycelium, Leptotrichia leptotrichia, degenerative forms of T. vaginalis, intra- and extracellular diplococci morphologically similar to N. gonorrhoeae, atypical cells or suspected atypical cells. Male reproductive function is examined by a combination of the semiological diagnosis and the obtained additional parameters of the ejaculate.

EFFECT: invention provides higher information value and reliability of diagnosis due to wider range of investigation of reproductive dysfunction factors.
