METHOD OF PRODUCING A HYBRID PREPARATION OF BROMELAIN AND CHITOSAN ASCORBATE IN FORM OF A THICK SOLUTION Russian patent published in 2024 - IPC C12N11/10 C12N9/50 

FIELD: biotechnology.

SUBSTANCE: disclosed is a method of producing a hybrid preparation of bromelain and chitosan ascorbate in form of a thick solution. Bromelain is immobilized with constant stirring at speed of 250 rpm by complexation in a thick solution of chitosan ascorbate in ratio of 10 ml of bromelain solution at concentration of 26 mg/ml, per 1 g of chitosan ascorbate with a molecular weight of 350 kDa, previously dissolved in 10 ml of a buffer. Formed preparation in the form of a thick solution is washed by dialysis against 50 mM tris-HCl buffer with pH 7.5. For washing with 21 ml of the obtained preparation, 200 ml of tris-HCl buffer is used. Dialysis is carried out for 8 hours, after which the buffer is changed to a portion of the buffer in volume of 200 ml. Dialysis is continued for another 16 hours until there is no free bromelain in the washing solution.

EFFECT: invention enables to obtain a hybrid preparation of immobilized bromelain and chitosan ascorbate in the form of a thick solution with viscosity in range of 200–250 MPa×s, having stability at 37 °C, completely washed from non-immobilized form.
