FIELD: bioengineering.
SUBSTANCE: invention relates to bioengineering, namely to a method for detecting regulatory elements of the genome, affecting transcription, and their use in biotechnologies. Mass parallel reporter analysis is used to assess the effect of one or another element of the genome on the expression of the target gene in the analysed cells. An important feature of the method is its high efficiency under conditions of processing a large amount of data. Due to the structure of the genetic reporter construct for creating DNA libraries used in the disclosed method, candidate DNA sequence in its composition is capable of affecting mRNA transcription from its own sequence, in case of presence of regulatory activity, which enables to implement the declared method.
EFFECT: disclosed method enables mass detection of endogenous regulatory elements of the genome, which affect transcription, to improve the characteristics of the protein accumulation process.
