FIELD: biotechnology.

SUBSTANCE: invention relates to a method of producing a bispecific antigen-binding polypeptide structure. Said structure comprises first (H1L1) and second (H2L2) heterodimers, wherein the first heterodimer comprises a first Fab region that specifically binds to the first antigen; and the second heterodimer (H2L2) comprises a second Fab region which specifically binds to a second antigen. Disclosed method comprises the following steps: 1) introduction of one or more substitutions into polynucleotides coding H1, H2, L1, L2; 2) introduction of said polynucleotides into a host cell and its cultivation; 3) collecting a bispecific antigen-binding polypeptide structure.

EFFECT: present invention provides antigen-binding polypeptides containing a light chain of immunoglobulin lambda and a light chain of immunoglobulin kappa, obtained using complementary amino acid modifications.
