SUBSTANCE: group of inventions relates to genetic engineering. Disclosed is a bicistronic plasmid construct FCU1-BsFm, a method for preparing the anti-tumour preparation FCU1-BsFm/PP, method of its delivery, methods of tumour cell replication inhibition and treatment of malignant tumours. Plasmid construct FCU1-BsFm encodes a chimeric gene of yeast cytosine deaminase and uracilphosphoribosyltransferase and a gene for secreted mutant flagellin Bacillus subtilis. To obtain a preparation FCU1-BsFm/PP, the plasmid structure is “packaged” in a carrier based on polyethylene glycol, polyethyleneimine and TAT-peptide. Method of treating malignant tumours involves intratumoural administration of the preparation FCU1-BsFm/PP in combination with the introduction of prodrug 5-fluorocytosine. Expression of the FCU1-BsFm construct in the tumour focus provides inhibition of tumour cell replication. Tumours represent sarcoma, colon tumour, lung adenocarcinoma, cervical cancer, breast cancer, ovarian adenocarcinoma or squamous cell carcinoma of skin.

EFFECT: inventions provide an effective method of delivering plasmid DNA into a tumour focus and inside cells, provide an anti-tumour effect mediated through a cytotoxic and immunostimulating effect.
