OPTIMIZED RELEASE OF ENDOGENOUS BETA-DEFENSINS Russian patent published in 2024 - IPC C07K1/14 C07K1/16 C07K1/36

Abstract RU 2825572 C1

FIELD: medicine; biotechnology.

SUBSTANCE: invention relates to medical biotechnology, specifically to release of endogenous beta-defensins from placental tissue. Disclosed is a method of separating beta-defensins from placental tissue cells, involving grinding, clarification with hydrogen peroxide solution, homogenisation in three steps: mechanical grinding, ultrasonic action with diameter of used nozzle 10 mm and increase of rotation speed to 2,000 rpm, time up to 30 minutes at temperature 4 °C, enzymatic hydrolysis of placental tissue cells with 2% trypsin on a magnetic stirrer, correction of the medium with Versene solution, cell sedimentation by centrifugation at 10,000 g, extraction of peptides on a separation column, with pre-ionisation of Sephadex G-25 by passing through the plates of direct current with force of 5 mA while passing the hydrolyzate through the Sephadex G-25 layer, followed by washing out beta-defensins from Sephadex G-25 particles with a cold Hanks solution, sterilizing filtration through fine-pore filters with pore diameter of 0.2 mcm and virus inactivation with acetic acid.

EFFECT: invention provides optimizing the recovery of the fraction of endogenous beta-defensins with high biological value for further development of pharmaceutical compositions.
