SUBSTANCE: present invention relates to molecular cytogenetics. Described is a method of producing a panel of DNA probes for determining chromosomal translocations, deletions and amplifications, involving development of a panel of DNA probes using long ones up to 15 kbp sequences of PCR amplifications with subsequent formation of DNA libraries for diagnosed sections of human genome and control loci, wherein said method includes steps of: selecting target regions of the genome; selection and synthesis of oligonucleotide primers for target regions of the genome; production of DNA products using PCR of long fragments; obtaining a library of DNA fragments using an enzymatic fragmentation method; ligation of the synthesized adapter sequences and amplification of the library using the synthesized target primer sequences; introduction into a genomic library of a fluorescent label; checking the specificity and intensity of the DNA probe in the FISH reaction.
EFFECT: disclosed method can be used to determine chromosomal translocations, deletions and amplifications based on fluorescent hybridization of a DNA probe with a genome region with a length of more than 30 kbp, in various biological samples of human cells.
