FIELD: biotechnology.
SUBSTANCE: invention relates to biotechnology, specifically to a method of producing a cell expressing a chimeric antigen receptor (CAR), comprising bringing a first target nucleic acid containing a TRAC sequence, in a cell in contact with a first nucleic acid/protein CRISPR composition containing a CRISPR targeting molecule and a Cas12 protein, and bringing a second target nucleic acid sequence comprising a B2M sequence in the same cell into contact with a second nucleic acid/CRISPR protein composition, containing a CRISPR guide molecule and a Cas12 protein, as well as to a CAR-expressing cell obtained using said method.
EFFECT: invention is effective for producing a CAR-expressing cell for adaptive cell therapy.
