FIELD: medicine.

SUBSTANCE: invention refers to medicine, namely to a method of extracting a stromal-vascular fraction from adipose tissue in test tubes in a sterile box. First, syringes with adipose tissue received in a laboratory are treated with 70% ethyl alcohol. Then adipose tissue is decanted in syringes. System for washing and incubation of adipose tissue is prepared – a container is taken to be attached to a container with blood components obtained after centrifugation, a plastic needle is removed at the end of the container line, 20 ml syringe without a piston is inserted into the obtained hole at the end of the container line. Then decanted adipose tissue is transferred into container without upper layer of adipose tissue. Further, the adipose tissue is washed twice with Ringer's solution at ratio of 1:1 to the adipose tissue and stirred in the container. Then it is held until the fractions are separated, the lower liquid fraction is removed through the main line back into the syringe, then 0.1% collagenase II solution is added to the washed adipose tissue at ratio of 1:1 to the adipose tissue, after which the main line is closed with a plastic clamp. Mixture of adipose tissue and collagenase is incubated in an incubator shaker at 37 °C for 30 minutes, after incubation , the cell suspension is diluted in the container with Ringer's solution at ratio of 1:1, wherein Ringer's solution is introduced through the container main line, after which the cell suspension is transferred from the container into the test tubes with a sieve, then the test tubes with a sieve and a cell suspension are centrifuged for 7 minutes at 400g in a centrifuge. After centrifugation, supernatant is removed from test tubes with a sieve by draining through the edge of the test tube, after which Ringer's solution is added to the test tubes with a sieve, and test tubes with a sieve and a cell suspension are centrifuged for 7 minutes at 400g in a centrifuge. After centrifugation, the supernatant is removed from the test tubes with a sieve by draining through the edge of the test tube. Then the bottom of the test tubes is unscrewed, the cell sediment is resuspended with a sterile pipette and the cell suspension is transferred into a clean test tube, thereafter, an aliquot is taken with an automatic dispenser for counting cells in the obtained stromal-vascular fraction, and the cells are counted on an automatic counter.

EFFECT: invention provides extraction of stromal-vascular fraction from adipose tissue in test tubes in sterile box conditions.
