FIELD: biology.
SUBSTANCE: invention relates to biology, specifically to a method for high-throughput DNA extraction from soya seedlings in 96-well plate. All steps of extraction are carried out in test tubes with strips in 96-well plate. Homogenisation of samples is carried out without addition of lysing buffer in several approaches, first without water, then with water (270 mcl per one sample). 2% CTAB lysis buffer is added immediately before thermostating (370 mcl per one sample). Further, the mixture is thermostated for 45-60 minutes. It is purified with a mixture of chloroform and isoamyl alcohol (24:1) in amount of 400 mcl per one sample. Centrifuged. 550 mcl of the upper layer is collected. DNA is precipitated with isopropyl alcohol in amount of 630 mcl per one sample. Centrifuged, supernatant is drained. DNA precipitate is washed once with 70% ethanol, dried and dissolved in water.
EFFECT: invention makes it possible to increase efficiency of DNA extraction of required purity and concentration for further molecular-genetic manipulations, such as, for example, PCR, and significantly reduce probability of cross-contamination of samples.
