FIELD: animal breeding.
SUBSTANCE: invention relates to poultry farming, namely to determination of the degree of morphological maturity of skeletal muscles of broiler chickens on 29th day of post-embryo embryogenesis on the basis of micromorphometric values. Method for assessing morphological maturity of skeletal muscles of broiler chickens consists in the fact that samples of skeletal muscle tissue of broiler chickens for histological studies are taken within 20 minutes after slaughter. It is fixed in 10% solution of neutral formalin for 24 hours. After washing in running water, muscle samples are dehydrated in dehydrating solution "Isoprep" and embedded in paraffin. Histological sections are prepared using LEICA RM 2235 microtome. Sections with thickness of 4-5 mcm are stained with haematoxylin and eosin. Micropreparations are studied using Microscreen and Jenamed-2 microscopes. During the light-optical examination of histological preparations, micromorphometry is carried out, followed by calculating a morphological maturity coefficient, which is a ratio of the thickness of muscle fibres multiplied by the number of muscle fibres in the field of view of the light microscope, divided by the product of thickness of endomysium and perimysium. Calculated value from 64 to 89 indicates presence in bird of low coefficient of morphological maturity, from 89 to 114 – average coefficient of morphological maturity and from 114 and higher – high coefficient of morphological maturity.
EFFECT: reliable determination of the structural state of skeletal muscles, which are significant for food consumption of the population, as well as manufacturability, simplicity, high accuracy of measurement with minimum time of analysis.
