FIELD: veterinary science.

SUBSTANCE: invention refers to veterinary virology. Described is a method for preparing a sample for isolating rabies virus in white mice, involving sampling tissues from the brain of white mice after euthanasia. This is followed by grinding tissues to size of 0.5–1.0 cm, centrifuging for 5–10 minutes at rate of 2000 rpm, introducing a daily culture into a growth medium with 10% embryonic blood serum of cattle and 3%, glutamine to concentration of 2 × 105 cells/cm3. Then the obtained suspension is added to the culture glass wells and placed in a humid CO2 incubator with CO2-content of 5% at temperature of 36–38 °C for 18–20 hours. Method is characterized by that the selected tissues after grinding are treated with 0.25% solution of trypsin for 10 minutes at temperature of 37 °C, and the growth medium used is the IGLA-MEM medium, into which the buffer agent HEPES is added in an amount of not more than 1% of the weight of the growth medium and the antibiotics penicillin and streptomycin at 100 units/cm3 and 1 mg/cm3, respectively.

EFFECT: obtaining reliable information in real-time diagnostics due to reduced risk of contamination.
