METHOD OF PRODUCING SQUALENE FROM SACCHAROMYCES CEREVISIAE “ВГШ-2” YEAST Russian patent published in 2025 - IPC C12P5/02

Abstract RU 2841142 C1

FIELD: biotechnology.

SUBSTANCE: invention relates to biotechnology and is a method of producing squalene from Saccharomyces cerevisiae “ВГШ-2” yeast, characterized by that yeast cultivation is carried out on amaranth flour hydrolysate, obtained by successive hydrolysis of amaranth flour preliminarily dissolved in distilled water with water duty 1:8, by enzyme preparations Amylosubtilin G3x at temperature 80 °C for 60 minutes and enzyme preparation dosage of 0.1 g per 100 g of flour and Glucavamorin G3x enzyme preparation at temperature 59-60 °C for 40 minutes and enzyme preparation dosage of 0.2 g per 100 g of flour, further, to stop the hydrolysis process, the mixture is added with glacial acetic acid with a weight ratio of 40% in amount of 1 ml per 100 ml of the hydrolysate, yeast cultivation is carried out under aerobic conditions on a medium of amaranth hydrolysate with addition of sugars, such as saccharose, glucose or maltose, in amount of 10 g per 1 dm3 of amaranth flour hydrolysate and terbinafine in amount of 0.3 mcmol per 1 cm3 of hydrolysate at pH=5.5 and temperature 32 °C for 48 hours, then the resulting yeast culture is separated from the medium by centrifugation for 5 minutes at a rotation speed of 4,000 rpm, then obtained biomass of yeast is subjected to autolysis by adding 75 cm3 of 96 wt.% ethyl alcohol solution and 50 cm3 60 wt.% potassium hydroxide solution per 1 g of absolutely dry yeast substance, autolysis is performed at temperature 55 °C for 18 hours, then squalene is extracted from the yeast autolysate with chemically pure hexane using 100 cm3 per 1 g of absolutely dry yeast substance, wherein extraction is carried out three times, further, hexane is evaporated from the extract in a vacuum evaporation apparatus, as a result, squalene is obtained with a degree of purity of not less than 98% and output of not less than 42 mg/dm3 of the nutrient medium.

EFFECT: invention enables to obtain squalene using an environmentally friendly method using a natural culture medium.
