INFLUENZA VIRUS STRAIN A (H3N2) FOR PREPARING OF LIVING INFLUENZA INTRANASAL VACCINE FOR ADULT AND CHILDREN Russian patent published in 1995 - IPC

FIELD: medicinal virology. SUBSTANCE: vaccine strain A/3/17/89 is a recombinant prepared by breeding of epidemic virus A/Transcarpathian region/354/89 (H3N2) with cold-adopted temperature-sensitive strain A/Leningrad/134/57/17 (H2N2) - an attenuation donor which is harmless for adult human and children. Strain A/3/17/89 is actively multiplied in developed chicken embryos at optimal temperature (32 C), difference of infectious activity indices at 32 C and 40 C is 6.75 lg ЭИД 50/0.2 ml. Recombinant inherited from actual epidemic virus 2 genes encoding surface proteins (hemagglutinin and neuraminidase), and 6 genes encoding internal proteins from attenuation donor. On the basis of data of recombinant-complementation analysis in genome of strain A/3/17/89 there are 3 ts-mutations that are typical for attenuation donor A/Leningrad/134/57/17 (H2N2). Strain A/3/17/89 is nonreactogenic for adult and children at the age 3-15 years at intranasal administration and causes 4-fold blood antibody increment in 60.0% adult and 73.0% children after a single vaccination. Strain is deposited in ГКВ at number 2275. EFFECT: increased effectiveness if strain indicated above.
