FIELD: biotechnology, genetic engineering. SUBSTANCE: invention involves the development of recombinant plasmid DNA pGSDE1 encoding the envelope protein E of tick-borne encephalitis virus and the strain of Escherichia coli as a producer of protein E. Recombinant plasmid DNA pGSDE1 is constructed on the basis of HindIII-ScaI-fragment of plasmid pEZZT 318 (size is 1165 base pairs), ScaI-SacI-fragment of plasmid pGEM1 (size is 1640 base pairs), SacI-BglII-synthetic fragment (size is 12 base pairs) and BglII-HindIII-fragment obtained by method of polymerase chain reaction (size is 1250 base pairs) that contains starting ATG-codon, encoding site of gene for protein E of tick-borne encephalitis virus and terminating TAA-codon also. The strain Escherichia coli GSDE - a producer of protein E of tick-borne encephalitis virus is obtained by transformation of plasmid DNA pGSDE1 carrying the gene E of tick-borne encephalitis virus. Recombinant protein can be used as an antigen in diagnostic test-systems in microbiological industry and medicine for development of highly effective prophylactic and diagnostic agents for control of tick-borne encephalitis virus.
