METHOD OF QUANTITATIVE ELECTROCHEMICAL ANALYSIS OF BIOLOGICAL MOLECULES Russian patent published in 2001 - IPC

FIELD: laboratory examination and analysis of various biological and natural liquids conducted in interests of medical diagnostics, commercial and ecological monitoring. SUBSTANCE: method includes formation of biological sensor by way of deposition of film of current-conducting polymer on surface of potentiometric electrode in process of electrochemical synthesis from monomer solution, realization of contact of biological sensor with sample of examined liquid, formation of electrochemical measurement cell from biological sensor and comparison electrode submerged in buffer solution and coupled by measurement device, recording of potential of biological sensor with regard to comparison electrode in working buffer solution with the aid of measurement device, recording of change of potential of biological sensor with sudden change of ion force of working buffer solution with constant value of its pH by means of measurement device and computation of quantitative content of analyzed component in examined liquid by value of change of potential of biological sensor. In this case current- conducting polymer is doped with anions showing highest capability for ion exchange with solution surrounding polymer at stage of electrochemical synthesis and avidin or streptoavidin is immobilized in or on film of current-conducting polymer. Biotic affine bioreceptors are used for immobilization of current-conducting polymer on film, they are used for realization of contact of biological sensor before formation of electrochemical cell. Contact of biological sensor with additional labeled bioreceptors is carried out prior to measurement of potential of biological sensor in working solution and change of potential of biological sensor is recorded with change of content of working solution. Method makes it possible to analyze small and uncharged biological molecules and to obtain strict quantitative results. It also allows storage time of realization of biological sensors to be prolonged, productivity and sensitivity of analysis as well as authenticity and reproducibility of obtained results to be raised.
