SUBSTANCE: method of polypeptide producing in yeast involves culturing yeast cells that are able to express this polypeptide transformed with yeast expression vector containing expressing cassette. Cassette has promoter sequence, DNA sequence encoding signal peptide, terminating sequence, DNA encoding peptide to be produced, DNA encoding processing site and DNA sequence LS encoding leader peptide of the general formula: Gln-Pro-Ile-(Asp/Glu)-(Asp/Glu)- X¹-(Glu/Asp)-X²-Asn-Z-(Thr/Ser)-X³ (I) where X¹ means peptide bond or encoded amino acid; X² means peptide bond or encoding amino acid or sequence involving up to 4 similar or distinct encoded amino acids; Z means encoded amino acid except for Pro and X³ means sequence involving from 4 to 30 similar of distinct encoded amino acids. Culturing is carried out in suitable medium up to attainment of expression and secretion of indicated polypeptide followed by isolation of polypeptide from medium.
