METHOD OF ASSAY OF HUMAN CYTOKINE STATUS AT GENETIC LEVEL Russian patent published in 2002 - IPC

FIELD: medicine, molecular biology, immunology, microbiology, virology. SUBSTANCE: invention relates to estimation of transcription levels of activity of genes encoding interferons, interferon-dependent and proliferative cytokines. Method involves determination of messenger RNAs for cytokines based on combination of methods of reverse transcriptasepolymerase chain reaction with blot- and dot-hybridization. For detection of alternations in cytokine status the following indices are studied: spectrum of mRNA for interferon-dependent and proliferative cytokines, namely interfrons (alpha-1, beta-1 and gamma), interferon-regulated enzymes: 2',5'- oligoadenylatesynthetase, RNAase L, ds-proteinkinase, factors of cellular proliferation: Bcl-2, Fas-antigen and protein of cytosceleton gamma-actin using synthetic oligonucleotides. Use of this method in clinical studies gives the necessary information concerning existing interrelationships in region of interferon-dependent cytokine reactions in cells at different pathological states.
