SUBSTANCE: invention relates to method for preparing hepatitis B surface antigen from recombinant yeast cells Hansenula polymorpha and can be used in biotechnology for preparing vaccine against hepatitis B. Method involves culturing recombinant yeast cells, disruption of cells in buffer containing chaotropic agent, addition of surface-active substance to cell homogenate, centrifugation of cell homogenate followed by hydrophobic chromatography and ultracentrifugation in potassium bromide density gradient, gel-filtration of fraction containing surface-active antigen. Hydrophobic chromatography is carried out in buffer containing 8-14% of ethanol followed by washing out with the same buffer at temperature 30 C and elution with the same buffer at temperature 40-42 C and gelfiltration is carried out on copolymer carrier. The preparation of surface antigen rHBs obtained from recombinant yeast cells Hansenula polymorpha is used for preparing vaccines for immunization against hepatitis B.
